利用药剂迅速把细胞杀死,以保持材料本来的状态和结构。
常用的固定有F.A.A固定液、卡诺固定液等。将固定液放在具塞小瓶中,投入材料,盖好瓶盖。用F.A.A固定液固定,时间至少24小时。F.A.A固定液既是良好的固定剂,也是保存剂,因此材料可长期保存在固定液中备用。卡诺固定液穿透力强,固定较小材料一般1~6小时即可,zui多不超过24小时。因卡诺固定液不能做保存剂,所以固定后要用95%、85%酒精浸洗,每级约20min.,然后保存在70%的酒精中备用。
(三)脱水
脱水的目的是除去组织中的水分,便于透明、浸蜡、包埋等步骤的进行,因为这些程序中所用的药品都不能与水混合,所以必须脱水。常用的脱水剂为酒精。脱水过程应缓慢进行,不能操之过急或时间过长,以免组织变硬变脆或发生收缩现象。脱水采用等级脱水(系列脱水),即从较低浓度酒精开始,逐渐替换到高浓度酒精。一般为:50%→70%→80%→95%(此时可加少许番红,使组织着色便于包埋时定位)→*(两次),每级停留时间2~4小时或更长(*中时间宜短)。
用95%酒精配制各浓度酒精的方法:设原有酒精浓度为A,所需配制酒精浓度为B,则取Bml原有酒精,加上A-Bml蒸馏水,即可获得Aml的B%的究竟。例如:95%酒精配成70%的酒精,则取70ml的95%酒精,加上25ml蒸馏水,则配成了95ml的70%的酒精。
